WALTHAM, Mass. – November 4, 2026 – Kailera Therapeutics, Inc. (Nasdaq: KLRA) (Kailera), a clinical-stage biotechnology company focused on elevating the next era of obesity care, today reported financial results for the third quarter ended September 30, 2026, and provided a summary of recent business highlights.

Kailera is advancing a diversified GLP-1-based pipeline of clinical-stage therapeutic candidates for the treatment of obesity. The company’s approach seeks to improve upon currently available treatments by developing products that have the potential to maximize weight loss, improve tolerability, and address other critical needs in the current therapeutic landscape. The Kailera portfolio includes single-, dual-, and triple-agonist mechanisms and both oral and injectable formulations.

“Kailera was built to bring new medicines to people living with obesity, and our strong execution this quarter reflects that commitment,” said Ron Renaud, President and Chief Executive Officer of Kailera. “All four of our product candidates are on track to deliver important clinical milestones in 2027, including the initiation of our Phase 3 trial for ribupatide oral and the data readout from the ribupatide injection high-dose Phase 2b trial. With the ribupatide franchise’s category-leading potential at the core of a diversified portfolio, and a balance sheet that supports multiple value-creating inflection points into mid-2028, I am proud of our team and energized by what we aim to deliver for people living with obesity."

Third Quarter Business Highlights and Recent Developments

The program is evaluating once-weekly ribupatide injection doses of up to 10 mg over 76 weeks in adults living with obesity or overweight. Data are anticipated in mid-2028.

•

Phase 2b high dose clinical trial in obesity fully enrolled with data anticipated mid-2027: The double-blind, randomized, placebo-controlled Phase 2b trial (NCT07458269)

enrolled approximately 260 participants in the U.S. with a BMI of 35 or greater without type 2 diabetes (T2D). The trial is evaluating once-weekly ribupatide injection doses of 10 mg, 13 mg, 16 mg, and 20 mg over 48 weeks. Data are anticipated in mid-2027.

The Phase 3 program will include two trials evaluating once-daily ribupatide oral doses up to 25 mg over 60 weeks in adults living with obesity or overweight with comorbidities, with and without T2D.

•

Global Phase 2 trial in obesity fully enrolled with data now anticipated in mid-2027: The double-blind, randomized, placebo-controlled Phase 2 trial (NCT07497880) enrolled approximately 300 adult participants in the U.S. and Australia with a BMI of 30 or greater or a BMI of 27 or greater with at least one co-morbidity, which may include T2D. This trial is designed to further optimize safiglipron's clinical profile for the treatment of obesity by evaluating a wide range of doses over 44 weeks, with a starting dose of 15 mg, and following a gradual, stepwise titration schedule designed to improve tolerability and determine the optimal balance between weight loss and tolerability. Data are anticipated in mid-2027.

Third Quarter 2026 Financial Results

As of September 30, 2026, Kailera had cash, cash equivalents and marketable securities of $1,037.6 million, which is expected to fund operations into mid-2028.

R&D expenses were $84.7 million for the quarter ended September 30, 2026, compared to $27.0 million for the same period in 2025. The increase of $57.7 million was primarily driven by the increase in product candidate development and the related nonclinical, clinical, and contract manufacturing costs associated with the Company's portfolio, as well as the increase in personnel-related costs to support its ongoing R&D activities.

General and administrative expenses were $21.7 million for the quarter ended September 30, 2026, compared to $13.1 million for the same period in 2025. The increase of $8.6 million was primarily driven by personnel-related costs and professional service costs as the Company continues to expand its operations.

Net loss was $95.7 million for the quarter ended September 30, 2026, compared to a net loss of $39.6 million for the same period in 2025.
